Most of the colleges ask for recommendation from current supervisor. Does it need to be necessarily my immediate manager. Can i take reco from my manager's manager. I was under my manager's manager for 2.5 years and for the past 1 year i have been working under my current manager. Will this require any additional information to be provided (in the optional essays) as to why i did this?

The person who knows you better and can share the strongest examples of your leadership and impact is the ideal recommender. Usually, your current role is the one in which you are making the most impact and for that reason it makes sense to use your current manager to write your recommendation. If your stronger leadership examples are from the previous role, then yes, the previous manager is a good choice. I do think it is worth a couple of lines of explanation in the optional essay or additional information space since the schools typically do request a letter from your current manager.
