does "the difference between A and B " mean "A-B" ???

Or we can't know which one minus the other one, it can be A-B or B-A????

Difference just means Greater number - Smaller number. Difference between two numbers is positive (or zero). Or, as you stated, we cannot know whether it is A - B or B - A if just the difference between them is given.

Difference between 3 and 8 is 5.
Difference between 8 and 3 is also, obviously 5.
