I'm thinking (A)

the word "that" should be used for clauses that are essential to the sentence, but here it introduces modifiers that are non essential.

Ill go with B. I agree that "that" is needed here. I like as instead of with. Not sure though.

My answer - B

You need the "that" (eliminates A and E)
D changes the meaning (eliminated)

Between B and C I went for B, I found the C incorrect. The with just doesn't sound right (don't ask me the grammatical reason why..)

can someone please throw some light on the usage of comma before such as ?

P.S. I am convinced with the OA for this question. Just want to understand the rule for "comma + such as" usage ?

It has to be B.

"That" is needed here. So, we are left with B,C, and D. Use of "while" makes D wrong. We are left with B & C.

It we leave out some of the words and only concentrate on the "soul" of the sentence, we get:

B --> As large.. companies.. that once did X... have downsized (As large companies...have downsized)

C --> with large.. companies.. that once did X.. have downsized (With large companies have downsized)

So, B makes more sense, making it the right choice.

Will go with (C). as the change has happened WITH some development.

I doubt if there will be any OA as the question was posted way back in 2008
