Statement 1: 5 is a prime factor of p.
This means p= 5*(something)
This does not confirm the # of 5s in p. Also, we are not sure if 2 is a prime factor of p.
Insufficient

Statement 2: 2 is NOT a prime factor of p. This means the positive integer p will not have any trailing zeroes.

B is sufficient

We can find the number of trailing zeros p has by dividing by powers of 5 until 5^k<p. How can we find the number of trailing zeros in p only by knowing the 2 is not a prime factor of p?
