What is known?, the number? or humans?
In this case, the number is known. In the sense, ABC are out.
Between D and E, I go for E.

"humans" in a passive form in A, B and C are not correct to modify the plant and animal species. D sounds awkward too.

Even though it is a little awkward, I think A is right too. E changes the meaning slightly and is passive.

The number of ... is over 800 - main clause
the underlined part following the relative pronoun that has no flaws

Rearranging the words for better understanding
(that) humans are known to --> (that) are known to humans which in turn act as a subject of the relative clause

the remaining sentences cite the reason for the extinction of species is ONLY humans

Answer A

Humans are known to have done something. Only A gets it right.
