1. 'and' means equal on either side. 50/50
2. 'as well as' means -- never 50/50; it may be 51 as well as 49 or 99 as well as 1 The first one is always more than the second in importance
3.In the case of 'not only'... 'bust also', the 'but also' factor is emphasized more than the 'not only' factor --e.g: Those who want to be employed in far eastern countries should be familiar not only with international etiquette but also with local customs. This means that if you don't regard local customs, you may be in for some trouble.

Now, in the given question, both strong boots and the backpack are equally essential for the hiker/ hikers. Yet again, the split on a large/ comfortable or comfortable/large backpack is just a diversionary ploy.

as well as is used when you are implying contrast to one thing compared to other.

Although "and" implies both items hold same importance.

But "as well as" could be used as "not only..., but also" - and such a meaning fits here well enough.
Consequently, B changes meaning...
While C implies the same meaning

The dilemma you are facing is chiefly because of your attempt to find the one correct choice that is mired with four other wrong choices. answer. This won't help in GMAT practice IMO; You should look forward to spotting errors first since there are four as many and it is easier to spot.
In this case, since I said both things are equal in importance, you must eliminate in one go A, C, and D, notwithstanding that there may be other errors in them. Then, on the glimpse of it, one can know that E is having an SV number error. B is the only one that is left.

The cornerstone of this confidence is the thoroughness of one's grammar basics.

Your Point no. 1 , 2 & 3 is very clear and help us to understand the usage of 'and; , 'as well as' and ' Not only... but also' . But in the present question or any other question, we can't understand the importance of 2 things from our own. We don't need to put any extra information from outside. We can't say if both the strong boots and backpack are equally important or not. If it is given in the question stem so we should not try to change the meaning.
And hence I feel that 'C' is a better option than 'B' for this question. I don't know the source but somehow the question is framed in a poor manner.

Are we at all concerned by the fact the OA says "Hikers need... a comfortable backpack"? Does it bother anyone but me that this seems to be implying many hikers to one backpack? And if not, why not?

Your confusion is not due to something you do not understand. Your confusion is due to the abysmal poor quality of the question. This is one of the worst SC practice questions I have ever seen.

The topic is 100% not something the GMAT would treat. Choice (E) has a SVA error, but essentially all four other answer choices are grammatically correct and acceptable: the differences among the answer choices are minute and not at all typical of the differences between answer choices on real SC questions on the GMAT. Other than awareness of the bare question format, it appears the question writer was utterly unfamiliar with the GMAT and its standards.

As someone who writes practice questions professionally, I would give this question a grade of an F.

I will attempt to be crystal clear:
Do not waste time analyzing atrocious questions: this analysis will not to prepare you for the GMAT.
If this question appeared in a print source, the very best thing you could do for your GMAT preparation would be to soak that print material in kerosene and set it on fire.

Explanation Why C is wrong :-

A Hiker needs strong boots

as well as

a large, comfortable backpack.

As well as :-
as said above is used to show that the first item is more necessary then the one
succeeding it and Gmat always tries to check the intelligence of the test taker I can comfortably assume
that they want us to make such assumption while answering this question.

Moreover we can find one more thing ,

A hiker --- is singular , so the coorect statement should be

A hiker needs strong pair of boots as well as a large , comfortable backpack

If you are not assuming that two items are equally important.

C and E are wrong because they both change the meaning of the sentence. Out of the remaining three B is the most concise, so we will go with it.

incorrect uses of "As well as".

It is not a synonym for 'and'.
Swedish is spoken in Sweden, as well as in parts of Finland. [incorrect]
This sentence means:
Swedish is spoken not only in parts of Finland, but also in Sweden. [Imagine! What a surprise!]
