I've heard that less traditional work experience is looked upon favorably in the b-school application process, but what exactly constitutes unique work experience? I've worked for two Fortune 100 retailers in Merchandising and Operations - would this be considered "unique?"

That's not unique, that's actually pretty standard.
Unique consists of things like administering vaccines in third world countries, working as an aide for the Obama administration, being in Teach for America, or managing a theater company -- things that aren't usually seen as "business" roles. It's especially impressive if their undergrad credentials COULD have landed them a prestigious standard job, but they chose to do the other thing instead. Working at an F100 company is highly corporate and businessy, and is the kind of employment you'd sort of expect in a business school class.
I wouldn't be too concerned with how unique your job is (after all, 30%+ of HBS students come from consulting or investment banking and it seemed to work out for them). Focus more on how you made a unique impact WITHIN a fairly standard job
