There are many benefits to timing your practice, including:
I'll be attending an applicant-initiated interview in December as I'm applying in Round 2. I've booked the Dartmouth Coach from NYC the day before and a room I found on Airbnb. Now, I'd really rather not wear my suit for a full 24 hours from early the day before, throughout the bus ride, and then in the morning for the interview for obvious reasons. However, I'll need to check out of my room before my interview and I'm unsure what I would do with a carry-on sized suitcase during the interview. I've read time and time again to avoid bringing a briefcase (to bring a padfolio instead), so I assume it would be frowned upon if I brought a suitcase!
I'll be hopping the Dartmouth Coach on the way out after the interview. I don't want to ask for special treatment or have anybody go out of their way to accommodate me - I don't want to rock the boat.
Any advice?

I believe that the interview is a whole day event, where you get to see the campus and also attend a class. Would you still be able to catch Dartmouth Coach in, let's say, late evening? Regardless, I'd advise you to not carry any such thing to interview. It appears to be rude and inconsiderate towards the host that you can't spare 2-3 days for such an important event.

Thanks for your response. My interview is on a Friday so I've taken that day and the day before off of work. The bus leaves at 2:30 p.m. and there isn't another bus until Saturday afternoon.

There you have it. Take the bus on saturday. Friday night you could spend with Tuckies. In fact, this can be your interview question for the interviewer! You could ask him if you may join any event taking place on Friday evening.
I like neo656's response. Additionally if you don't want to impose on your interviewer, you will meet other students throughout the day that can help. If it's a class day, you'll have a student escort. The tour is conducted by a friendly Tuckie. And the lunch Q&A has several more.
Also, there's plenty of coat space at the entrances (coming from Texas, this fascinated me that square footage is allocated for coats!). I'm sure you can leave a suitcase there or in the candidate waiting area, and the interviewer would never know.
Check in with admissions, they may have some place you can store it, or the area between Stell and Byrne, there is a coat area where you could leave a piece of luggage. It's not a big deal at all!
