Yes, it would matter and help, particularly if that upbringing still impacts who you are today and your accomplishments. For example, are you involved in any organizations that give back to rural, underdeveloped communities such as your own? Then, it would be more than appropriate for you to discuss your hometown. Without that context, your involvement would make less sense.
That said, tread lightly. The admissions committee will clearly pick up if you are trying to play the diversity or pity cards with an on-the-nose approach.
FWIW, the rural, underdeveloped community aspect sells way more than your family's education background, though I am sure the two are inextricably linked.
