Hi when we use IN / BY verbing then the subject is performing those actions.
Here spiel is not emphasizing on tiger welfare and conservation, the ringmaster is.
So we need something that would describe "spiel " which is what C is doing.

(i) ringmaster's spiel IS UNUSUAL IN ITS EMPHASIS on tiger welfare and conservation.
(ii) ringmaster's spiel IS UNUSUAL IN EMPHASIZING on tiger welfare and conservation.

(i) the spiel is unusual in its emphasis on....talks about the spiel's emphasis, well, ok.
(ii) the spiel is unusual in emphasizing... says "spiel" is emphasizing... oh, i can see the difference.

But besides, are there other errors?

how are you doing?
I would like to add one thing in yor post.
In "in its emphasis", how can "its" refer to a belonging of a human. If I am not wrong, if the ringmaster is emphasizing then it must be "his". Moreover, I really want to know the source of this question because IMO "it" is ambigous here.
