→ The sentence sets up a contrast between two things: the environmental consequences of engineered genes escaping into the wild, which are difficult to evaluate, and the odds of such escapes actually happening, which can be evaluated.
THE OPTIONS
The word since in this context means because and does not provide logical contrast.
• Illogical
The odds of escapes happening can be evaluated, but not because the environmental consequences of escaped genes is hard to evaluate.
ELIMINATE A
→ the word but provides the contrast needed
→ I do not see any errors.
KEEP
→ had escaped is past perfect, the "past of the past."
Almost always, in order to talk about the earlier of two past events by using past perfect, we need to see at least one past event that happened: a simple past tense verb, a time stamp, or another context that makes it clear that one event preceded the other.
The sentence is talking about a general truth, in which case present tense should be used.

→ The use of and fails to bring out the contrast inherent in the sentence.

My answer is (B). It took me 01:08.
(A) "Since" can mean "because" or "From the time when". GMAT tends to insist that "Since" be used with time.
But even if we interpret "Since" as "Because", the sentence still does not make sense.
(B) This coordinated sentence is straightforward. No drama. Clear meaning.
(C) "While" can mean "during the time that" or "although". GMAT tends to insist that "While" be used with time.
But even if we interpret "While" as "Although", the sentence is incorrect in the use of "that had escaped into the wild", for two reasons:
(C.1) "Past perfect tense" refers to some action that happened before another past event. There is no past event in the sentence. If anything, "had escaped" should be "escaped".
(C.2) Even if we change "had escaped" to "escaped", it acknowledges that such leak actually occurred. The meaning does not jibe with the main clause, which, by using "actually happening", indicates such occurrence may have not been confirmed.
(D) "and" does not indicate the correct relationship between two independent clauses.
