If you want to use the possesive ('s), the correct noun would be Virginia City's business.
I think that "in" or "of" are almost the same. Try to eliminate the sentences finding other errors.

I believe businesses in Virginia city will change the meaning as it only counts businesses inside the virginia city, whereas businesses of means businesses operated from Virginia city. regarding possessive, 'businesses of virginia city's' looks awkward. we generally say virginia city's something ....use of OF before Virginia city already tells that Virginia city possesses something.
