Most MBA programs have R1 deadlines around September and second deadlines are now mostly next year i.e. Jan 2022. My question is to understand that if one applies in round 2, does that in some way backfire the student? Is it a wise decision to apply in Round 1?

Not at all. Generally speaking, Rounds 1 and 2 are the same in terms of admission statistics and scholarship awards. You are at no disadvantage by waiting until the January 2022 deadlines to submit your application.
People wait until Round 2 because they aren't ready to apply in R1 for whatever reason (didn't decide to attend business school until later in the year, may need to retake the GMAT, recently switched jobs, etc.) The most important thing is to make sure you put the best application forward in whichever round you do apply.
