It is a good question but I agree with mangamma - you have to be more specific.
In any case, negotiating a job offer is best accomplished if you have a great alternative. Without one, it is pretty challenging.
Also, people negotiate multiple things - money, level, location etc. I recently went through this process and happy to chime in for specific questions!
