I was waitlisted in Round 1 and then subsequently waitlisted again in Round 2. What does this effectively mean and are my chances of admission practically zero now?

It's a bit rare. But it happens. It means that you are still good enough to consider, but that they are still holding out for something better. Maybe you should not give them the chance?

did you write them an essay or a letter emphasizing your desire to attend? Have you done anything since you got on the waitlist? What?

Yes, I've written them several letters already to express my desire to attend (+ updates at work). Do people who hear back after being waitlisted in Round 2 typically hear back during Round 3 decisions?

It depends, I've worked with a couple of people who got answer in AUGUST!!!! At the very tail end of results. Thankfully they got in. :)

But just keep doing what you are doing then: Let them know you are alive and interested. And if it begins to drag out a bit long, come up with a backup plan (like retake GMAT or whatever for next season).
