From people I have talked to, it should generally be from people that you work with on a consistent basis who can speak to your candidacy. Further, it helps if they are an MBA (especially a Columbia MBA), as they can better attest to why an MBA is right for you, why Columbia (one of the questions they ask on the recommendation form), and your leadership potential.
If you spend a significant portion of your time in your extra-curricular activity (and it shines through in the rest of your application) and the person can go in depth into your ability to lead, be an MBA, etc. then it doesn't hurt. If its something that you only do a couple hours a week (or less), you would probably be better off with a current manager at work.
Of course, it all depends. I'm guessing you're asking this question because there may not be anyone else capable of writing a great recommendation (happened to me since I'm at a tiny firm - ended up having a former boss write one). If that is the case, go with the senior person from your extra-curricular activity. Just make sure somewhere in your application, Columbia knows how much time you spent with this person. If you rarely see them, it might call into question how this person can truly assess your skills, no matter how good the recommendation is.
