Firstly, YLP is an entry only for candidates who are currently pursuing their final year of the undergraduate degree. From my understanding of the information that you have given, you have been working from December 2016. This automatically rules you out from applying through the YLP route. However, you can apply through the usual PGP route. The EEO route is for candidates who have less than 2 years of work experience as on 31st March 2019 (although I am not sure about the exact cut off, you fall under the PGP route since you complete 2 years in December 2018)
Now coming to the two stages of admissions, there is no specific distinction between the two rounds other than the reason that a few seats will get filled up in the first stage. So the earlier you apply the better. Now coming to GMAT, You can write it at any time of the year at your convenience. The only thing that ISB checks is whether your GMAT score was submitted before the respective stage of admission that you have applied for.
